Description générale

Xanthine Oxidase (XO) catalyzes the sequential oxidation of hypoxanthine to xanthine, and xanthine to uric acid and hydrogen peroxide. In humans and other primates, XO controls the final step of purine catabolism and is normally found in the liver and the intestinal mucosa. In rodents, XO is broadly expressed in most tissues. While XO activity is normally very low in blood, liver injury can result in the release of XO into blood. XO may contribute to the pathogenesis of gout and cardiovascular disease, and XO activity or expression may be upregulated in these conditions.

Principe

The Xanthine Oxidase Activity Assay Kit provides a simple and direct procedure for measuring XO activity in a variety of biological samples. XO activity is determined by a coupled enzyme assay, which results in a colorimetric (570 nm)/ fluorometric (λex = 535/λem = 587 nm) product, proportional to the hydrogen peroxide generated. One unit of XO is defined as the amount of enzyme that catalyzes the oxidation of xanthine, yielding 1.0 μmole of uric acid and hydrogen peroxide per minute at 25 °C.

  1. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   3. Does the kit allow the use of cell pellets that have been frozen at -80°C, or do the samples need to be fresh, considering that the cells should be rapidly homogenized with the assay buffer?

    1. The cell pellet can be stored at -80°C with a small amount of PBS, avoiding storage with culture medium. Upon thawing, it's recommended to wash the cells once with PBS and then remove the PBS. Following this step, the assay buffer should be used for extraction according to the kit instructions. It's important to avoid multiple freeze-thaw cycles. However, the supplier does not specify the maximum time frozen cells will remain stable for assay.
