Description générale
Leur configuration verticale et leur surface utile de filtration en font des dispositifs très performants : traitement rapide, très bon taux de récupération (en général plus de 90 % de la solution de départ diluée) et possibilité de concentrer l'échantillon 80 ×. Leur configuration verticale permet en outre de minimiser la polarisation du soluté, et donc, le colmatage de la membrane qui en découle. Enfin, la présence d'une butée physique à l'intérieur du dispositif empêche d'atteindre la siccité tout en prévenant les éventuelles pertes d'échantillon.
Le concentré est récupéré dans le réservoir à échantillon du filtre à l'aide d'une pipette, tandis que l'ultrafiltrat est recueilli dans le tube à centrifuger fourni. Le filtre peut être centrifugé dans un rotor à godets oscillants ou à angle fixe. Avec leur membrane verticale et leur butée fixe, les filtres à centrifuger Amicon® Ultra de 15 ml offrent un taux de récupération maximal pour un temps de centrifugation relativement court.

Hello, Are these tubes valid for AAV concentration? And if so, are these valid for all serotypes? Also, the user guide says that the membrane's material is 'Ultracel® low binding regenerated cellulose'. What's the difference with the PES membrane?

These devices can be used for virus concentration but are not validated for any specific type. Both PES and regenerated cellulose membranes have low binding, but PES has a faster flow rate. Hi, I have plan to use the Amicon Ultra-15 100K for Exosome extraction from cell culture supernatant, could you please let me know what would be the best speed (rpm/rcf) and time for doing that? Thanks

Typically, Amicon-15 devices are spun at 4,000 x g (relative centrifugal force) for exosome isolation. However, spin times may need to be optimized, based on the sample type and volume. What is the minimum starting volume I can load on Ultra 100K (100,000 MWCO) devices? Can I load as little as 500 µL?

This product is intended for use with 4-15 mL initial sample volumes. The device has not been evaluated for use with volumes smaller than those recommended. While it may be possible to concentrate samples as low as 500 uL, the recovery volume cannot be predicted. Does the Amicon Ultra has residues that may released into the concentrated product', like Beta-Glucan?

These devices contain trace amounts of glycerine. If this material interferes with downstream analysis, rinse the device with buffer or Milli-Q® water prior to use. Should interference continue, rinse with 0.1 N NaOH followed by a second spin of buffer or Milli-Q® water. NOTE: Do not allow the membrane to dry out following rinse steps. If the product will not be used immediately after rinsing, leave the final buffer or water on the membrane until the usage. Please see the link below to review the full product User Guide:

I just noticed that Amicon ultra - 15 centrifugal filters are non-sterile. Has this been always the case? REF: UFC910024, LOT:0000193199

The Amicon Ultra devices have always been non-sterile. These devices cannot be sterilized by UV or autoclaving. However, it is possible to sanitize the devices with 70% ethanol. Please see the protocol for sanitization below:
1. Reagent preparation: 70% Ethanol solution (v/v) is prepared by dilution in distilled water. Both the ethanol solution and distilled water used for rinsing should be filter-sterilized using sterile, 0.22 µm sterilizing grade filter devices, such as the 50 mL Steriflip® device SCGP00525 or SE1M179M6.
2.Sanitization Protocol:
A. Fill the Amicon® Ultra concentrator device to its published capacity with filter-sterilized 70% Ethanol, and cap the unit (note that the collection tubes can be sanitized in the same manner).
B. Invert device by hand or in an end-over-end mixer to continuously wet all internal surfaces of the device.
C. Incubate for a minimum of 5 minutes (note that the device can be capped and left for up to 24 hours if it is not going to be used immediately).
D. Spin at recommended G-force using appropriate centrifuge until at least 50% of the volume has passed through the device.
E. Aseptically discard any remaining ethanol from the filter device and filter collection tube.
F. Immediately rinse away residual ethanol by adding the filter-sterilized water to the device's published capacity, mix gently to wet all areas, and centrifuge until at least 50% of the volume has passed through the device.
G. Discard any remaining liquid from both the concentrator device and filter collection tube before use.DO NOT let the filter membrane dry out in concentrator unit. If the devices are not to be used immediately, fill the devices with filter-sterilized distilled water, cover, and store refrigerated. Remove liquid from device as previously described prior to use.

What is the maximum volume that can be loaded into the Amicon 15 device when using both fixed angle and swinging bucket rotor centrifuges?

When using the fixed angle centrifuges with the Amicon 15 device, the maximum volume that can be loaded is 12 mL. It is important to place the device at a 35-degree angle. On the other hand, when using swinging bucket centrifuges, the maximum volume that can be loaded into the Amicon 15 device is 15 mL.

Hii, I wold like use these tubes for ultrafiltration of FBS to deplete it from EVs. I found they were used in one publication for such an experiment. How can they be sterilized so they can be used in cell culture hoods?

Hi, Can you please confirm the centrifugation speeds ( in rcf ) for the Amicon-15 100kDa and the 10kDa filters ? I was told that the max speed for the 100kDa is 3000g and for the 10kDa is 4000g, respectively. Is that the case and if so, why exactly ?

Spin times range from 10 - 40 minutes and will depend on the sample types as well as the rotor (fixed-angle or swinging bucket). See the image below for spin times and speeds for each rotor type.
