General description

The UPC-10 tumor line that produces mouse IgG2a, K is a tetramethylpentadecane (pristane) induced plasmacytoma originated and carried intraperitoneally in BALB/c mice. The hapten binding specificity of the UPC-10 line is for β-2-6-linked fructosan. The ascites produced from the UPC-10 tumor line contains IgG2a, K in addition to normal levels of other mouse immunoglobulins and serum proteins. The ascites fluid is clarified by centrifugation and filtration. IgG2a (immunoglobulin G 2a) is one of the subclasses of rat immunoglobulins, which also include IgG1, IgG2b and IgG2c.

Specificity

Antigen Specificity: β-2,6-fructosan

Trace amounts of lambda chain may be present.

Biochem/physiol Actions

The IgG2a (immunoglobulin G 2a) plays a vital role in stimulating autoimmunity in mice. This gene is expressed at high level during virus infection. IgG2a offers host defence through activation of complement system. This antibody has specificity towards Fc receptors expressed on phagocytes and stimulates antibody-dependent cell mediated cytotoxicity. IgG2a also induces macrophage-mediated opsonophagocytosis, which has been observed in response to influenza virus and leads to its clearance from the infected cells.

Physical form

The product provided is lyophilized from 0.01 M phosphate buffered saline, pH 7.2, with no preservatives added.

Preparation Note

Storage and Stability: Store the product at 2-8 °C. After reconstitution, the solution may be stored frozen in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage clarify the solution by centrifugation before use.
