Description générale

VGLUT1 (vesicular glutamate transporter 1, originally termed BNPI and characterized as a Na+/Pi transporter) is a 60 kDa protein. VGLUT2 is enriched in synaptic vesicles and selective for a distinct class of glutaminergic nerve terminals.
Vesicular glutamate transporter 1 (VGLUT1), also called as solute carrier family 17 member 7 (SLC17A7), is encoded by the gene mapped to human chromosome 19q13, a region associated with schizophrenia. VGLUT1 is specifically expressed in neuronal cells in the brain and at higher levels in neuron-enriched regions such as the amygdala and hippocampus. Glia-enriched areas such as the corpus callosum also show moderate level of expression and substantia nigra, subthalamic nuclei and thalamus show low level expression. VGLUT1 is characterized with a conserved C-terminal dileucine-like motif and two polyproline domains distal to C- terminal end, including one that binds to endocytic BAR (Bin/Amphiphysin/Rvs) domain protein, endophilin.

Immunogène

synthetic peptide corresponding to a region near the C-terminus of human VGLUT1 (amino aicds 504-520).

Actions biochimiques/physiologiques

Vesicular glutamate transporter 1 (VGLUT1) is associated with neuronal pathways that exhibit activity-dependent potentiation, whereas VGLUT2 is expressed in sensory and autonomic pathways that display high fidelity neurotransmission.
Vesicular glutamate transporter 1 (VGLUT1) plays a vital role as a vesicular glutamate transporter in glutamatergic neurons. VGLUT1 expression in the prefrontal cortex functions as a potential neurochemical marker of dementia in Alzheimer′s disease (AD) and antidepressant activity.

Several lines of evidence suggest that the glutamatergic system is severely impaired in Alzheimer disease (AD). Here, we assessed the status of glutamatergic terminals in AD using the first available specific markers, the vesicular glutamate transporters VGLUT1 and VGLUT2. We

The two recently characterized vesicular glutamate transporters (VGLUT) presynaptically mark and differentiate two distinct excitatory neuronal populations and thus define a cortical and a subcortical glutamatergic system (VGLUT1 and VGLUT2 positive, respectively). These two systems might be differentially implicated in
