Description générale

Glycerol, also referred to as glycerin or glycerine, is a 3-carbon sugar alcohol that forms the backbone of fatty acids. It is also a component of phospholipids and triglycerides. Glycerol is a sweet-tasting, hygroscopic liquid. Glycerol is widely used in food, beverage, and pharmaceutical formulations[1] and is the main waste by-product of biodiesel production via transesterification.

Application

Glycerol assay kit has been used to quantify glycerol release from mouse 3T3-L1 cell culture supernatant samples[2] and to measure plasma glycerol levels.[3]

This kit is suitable for glycerol detection in foods, beverages, pharmaceutical formulations, and biological samples.

Principe

In this kit, glycerol concentration is determined by a coupled enzyme assay involving glycerol kinase and glycerol phosphate oxidase, resulting in a colorimetric (570 nm)/fluorometric (lex = 535/lem = 587 nm) product, proportional to the glycerol present. The linear range of detection for this kit is 10-1,000 μM (colorimetric) and 2-50 μM (fluorometric).

  1. After reading the description of this product, I can't help but wonder what the contents of the kit are, and if there is any additional documentation related (protocol example, enzyme description, standard concentrations and curves).

    1. Full product information, including the protocol, is available in the product datasheet. The formulation of the components is considered proprietary, with the exception of the standard - 100mM Glycerol. This is an enzymatic assay that utilizes glycerol kinase and glycerol phosphate oxidase. Please see the link below to review additional details:

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. Is there any information available on how to prepare tissue for assay, considering that technical bulletin MAK117 does not include any recommendations on this matter?

    1. The general recommendation for tissue sample preparation is as follows:
      1. Begin with 20-100 mg tissue and add 200-1000 uL ice-cold PBS.
      2. Lysis can be achieved by homogenization (10-20 passes in a Dounce homogenizer on ice) or by sonication (preferably performed in an ice-water bath). The degree of tissue lysis can be checked under a microscope.
      3. Centrifuge the homogenate at 14,000 g for 10 min.
      4. Transfer the clear supernatant into a clean tube and use it for the assay.

  4. Can serum glycerol content be analyzed or just plasma?

    1. Yes, this product can determine glycerol content in serum samples.

  5. How long are reagents stable at 2-8 C after initial thaw?

    1. The Assay Buffer, MAK117A, may be stored at 2–8 Celsius. For all other components, it is recommended to aliquot and freeze at - 20 deg C. Avoid repleated freeze/thaw cycles.
