Description générale

L'hydroxyproline (4-hydroxyproline) est un acide aminé non protéinogène[1] formé par l'hydroxylation post-traductionnelle de la proline. L'hydroxyproline est un constituant essentiel du collagène, dont elle stabilise la structure hélicoïdale.[2] Parce que l'hydroxyproline est essentiellement présente dans le collagène, son taux peut constituer un indicateur de la concentration en collagène. Les conditions qui favorisent le renouvellement du collagène peuvent accroître les taux d'hydroxyproline sérique et urinaire. Les taux d'hydroxyproline sérique et urinaire peuvent donc servir de marqueur de la résorption osseuse.[3][4]

Application

Le kit de dosage de l'hydroxyproline a été utilisé pour déterminer la concentration en hydroxyproline et en collagène.[5][6][7]

Adéquation

Utilisable pour la détection de l'hydroxyproline dans les surnageants de cultures cellulaires et tissulaires ainsi que dans les échantillons d'urine, de plasma, de sérum et d'autres échantillons biologiques.

Principe

Le kit de dosage de l'hydroxyproline permet de déterminer la concentration en hydroxyproline en faisant réagir l'hydroxyproline oxydée avec du 4-(diméthylamino)benzaldéhyde (DMAB), donnant un produit mesurable par colorimétrie (560 nm) dont la quantité est proportionnelle à la quantité d'hydroxyproline.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

    1. This type of sample has not been tested with this assay kit. It is suggested to consult the scientific literature to determine if the specific sample type has been reported for this type of usage.

  5. 1. Are heart samples that have been flash frozen suitable for use with MAK008-1KT?

    2. The manual mentions 10 mg of tissue, but does it need to be an exact measurement? Is it challenging to accurately weigh the frozen tissue?

    1. 1. Flash frozen tissue can be used with the kit. The recommended approach is to homogenize the tissue with room temperature water. Allow the tissue to thaw slightly by leaving it in the water for a few seconds before homogenization.

      2. The kit is optimized for use with 10 mg of tissue homogenate supernatant. Slight variations below or above 10 mg will not affect the assay significantly. However, it is advisable to avoid large differences in the weight or mass of the tissue.

  6. Is it possible to carry out the 90-minute incubation at 60 °C, as mentioned in the protocol after adding the DMAB Reagent, using a hot plate? and, should the plate be covered during this incubation step?
