General description

X-Gal is a chromogenic substrate for β-galactosidase that produces a rich blue color that can easily be detected visually over background. It is soluble in dimethylformamide and methanol. The product is not soluble in water.

Application

X-Gal has been used for immunohistocytochemistry applications. It is also the substrate of choice for blue-white selection of recombinant bacterial colonies with the lac+ genotype.

Preparation Note

Working solution: Stock solution:
  • 20 mg/ml in dimethyl formamide (DMF), clear, colorless solution
  • A590 nm: 0.01

Storage conditions (working solution): X-gal in solution (i.e., dissolved in DMF 2% (w/v)) is stable for several weeks when stored in the dark at 2 to 8 °C; when the solution is stored at -15 to -25 °C, it is stable for one year.
