Biochem/physiol Actions

Proteinase K is a stable and highly reactive serine protease. Evidence from crystal and molecular structure studies indicates the enzyme belongs to the subtilisin family with an active-site catalytic triad (Asp39-His69-Ser224). It is stable in a broad range of environments: pH, buffer salts, detergents (SDS), and temperature. In the presence of 0.1-0.5% SDS, proteinase K retains activity and will digest a variety of proteins and nucleases in DNA preparations without compromising the integrity of the isolated DNA.

One unit will hydrolyze urea-denatured hemoglobin to produce color equivalent to 1.0 μmole of tyrosine per min at pH 7.5 at 37 °C (color by Folin-Ciocalteu reagent).

  1. Hi, I'd like to know what happens if the lyophilized proteinase K (P238) is not stored at -20C for at least a week.

    1. The decline in activity over time due to storage conditions has not been determined. Other Proteinase K products may ship at ambient temperature, indicating little or no degradation of the material is expected. However, this items ships on ice. Maintaining the shipping and storage temperature at -20°C is recommended for this product in order to assure optimum performance.

  2. Is P2308 designed for antigen unmasking during IHC, and is there literature supporting this application?

    1. Proteinase K has not been tested for use with IHC. Product P2308 is specifically tested for use in molecular biology, particularly for preparing cell lysates and releasing nucleic acids, as well as for PCR applications. While the enzyme has been used to digest hair (keratin) and may have other applications, there are no guarantees that P2308 would be suitable for antigen retrieval. No literature references stating that Proteinase K is commonly used for IHC were found.

    1. This product may be dissolved in water for nucleic acid preparations at 1 mg/mL.

  4. How to prepare a solution of 20 mg/ml of pK (P2308-5MG)? In the section "Preparation Instructions" it mentions that I need to solve 1 mg in 1 ml of water. Does it means I'll have a final concentration of only 1 mg/ml?

    1. The stock may also be prepared at 20 mg/ml concentration in 50 mM Tris-HCl, pH 8.0, containing 1 mM calcium chloride. This solution is stable for at least 12 months at 4°C. For additional information, please refer to the below publications where the product P2308 has been used.

  5. How to prepare Proteinase K solution to use in antigen retrieval of formalin fixed paraffin embedded tissue sections for the purpose of paraffin immunoflorescence? Which is the diluent of choice to prepare the solution from powder form of proteinase K.

    1. This product is not tested for use in antigen retrieval. The product 21627, IHC Select Proteinase K protocol may be applied to this product, P2308. To prepare a stock solution, dissolve Proteinase K, P2308, at 0.2mg/mL in 0.05M Tris solution, pH 7.5.
      For antigen retrieval in FFPA tissue sections (deparaffinized) the recommended dilution of the stock solution is 1:30.

  8. How should I store solutions of Product P2308, Proteinase K?

    1. Stock solutions in 40% glycerol of at least 20 mg/ml can be stored in 10 mM Tris-HCl buffer, pH 7.5.with 1 mM  calcium acetate (see product P4850). Solutions are also stable at pH 8 at 4°C.
