General description

VU0360172 is a potent (EC50 = 16.6 nM), systemically active positive allosteric modulator (PAM) of the metabotropic glutamate receptor subtype 5 (mGlu5). It is the first orally active mGlu5 PAM to be efficacious in an in vivo preclinical antipsychotic model, producing a dose-dependent reversal of amphetamine-induced hyperlocomotion.[1] It was also found to reduce spontaneous absence seizures in WAG/Rij rats and to inhibit pain. Reduced spontaneous spike and wave discharges without impacting motor behavior in a rat model of absence epilepsy have been observed.
