Application

ALX 5407 hydrochloride has been used as a glycine transporter (GlyT1) inhibitor to test its effect on inhibitory postsynaptic current and N-methyl-D-aspartate receptor (NMDA)-mediated excitatory postsynaptic currents (EPSCs).[1] It has also been used as a GlyT1 inhibitor to treat developmentally diminished NMDA receptor (Grin1D481N) mice to test its effect on the glycine site function.[2]

Biochem/physiol Actions

ALX 5407 is a selective, irreversible GlyT-1 glycine transporter inhibitor.

ALX-5407 hydrochloride (NFPS hydrochloride) is a selective irreversible inhibitor of the glycine transporter GlyT1 with IC₅₀ values of 3 nM for GlyT1 compared to 100 μM for GlyT2. ALX-5407 hydrochloride showed no activity at the inhibitory glycine receptor or glycine site of the NMDA receptor (IC₅₀ > 100 mM).
