General description
Durch die Pufferkapazität von PBS, die durch ein Gleichgewicht zwischen Phosphat und Phosphorsäure vermittelt wird, wird die pH-Umgebung stabilisiert, was für eine optimale Enzymaktivität und verschiedene biologische Prozesse entscheidend ist. Dadurch ist PBS ideal für die Zubereitung von Reagenzien, für die Proteindialyse und die Immunhistochemie und gewährleistet konsistente und zuverlässige Versuchsergebnisse. Über die Zellbiologie hinaus findet PBS auch Anwendung zum Beispiel bei der Entwicklung individueller ELISA-Puffer für optimale Antikörper-Antigen-Interaktionen. When using in cell culture do you recommend autoclaving and filter sterilizing? Or only autoclave?

Phosphate buffer by itself is not generally autoclaved. When phosphate buffers are autoclaved, it is common to see a precipitate form at the bottom of the bottle. When phosphate buffers are a minor component in a solution to be autoclaved, the formation of precipitate does not seem to be a common occurrence. Instead of autoclaving the phosphate-buffered saline (PBS), sterilization can be achieved by passing the solution through a 0.2 micron vacuum filter. How can I determine the shelf life / expiration / retest date of this product?

If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.

The contents of each package are sufficient to prepare 1 Liter. To prepare, fill the vessel of choice (beaker, graduated cylinder, etc) to approximately 800 mL of distilled or deionized water. Place the vessel on a magnetic stir plate and add the appropriate-size stir bar. While stirring, add the contents of 1 package. Add the required additional volume of water to 1 liter. Allow the solution to stir until the contents are completely dissolved and no participates are present.

Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. The composition of P3813 PBS Phosphate Buffered Saline is considered proprietary, but the website provides the following information. When one pouch is dissolved in one liter of distilled or deionized water, it yields 0.01 M phosphate buffered saline with a pH of 7.4 at 25°C, consisting of NaCl at 0.138 M and KCl at 0.0027 M.

Is autoclaving necessary for PBS P3813 before conducting the presence-absence test for coliform bacteria?

No, phosphate buffers by themselves are generally not autoclaved due to the common occurrence of precipitate formation. Autoclaving is commonly used in microbiology, but if your test method only requires sterilization without specifying autoclaving, filter sterilization might be a better option to consider.

PKG means package. One package is one pouch. As per its COA: https://www.sigmaaldrich.com/certificates/sapfs/PROD/sap/certificate_pdfs/COFA/Q14/P3813-1PAK0000291289.pdf, each pouch contains 9.07-10.00 g of powder. And the pH and dissolution time are tested to validate the solution.
