Application
TLR3 erkennt Doppelstrang-RNA und ist ein wichtiger Effektor der Immunreaktionen gegen virale Erreger. Polyinosin-Polycytidyl-Säure (Poly(I) • Poly(C)) ist ein Doppelstrang-Homopolymer und wird als Modell-RNA zur Erforschung der Zellkommunikation auf TLR3-Ebene verwendet. Poly(IC) ist ein TRIF-abhängiger TLR3 (Toll-like-Rezeptor-3)-Ligand.
Biochem/physiol Actions
Die Transfektion von Poly (I:C) in NIT-1-Zellen wurde im Rahmen eines Modells für die intrazelluläre dsRNA-induzierte β-Zell-Apoptose verwendet. Achtzehn Stunden nach der Transfektion waren 45 % der Zellen apoptotisch und es wurde eine Zunahme der nukleären Translokation von NF-kB, p50/p65, die Spaltung der Caspasen 3 und 8 sowie die transkriptionale Induktion von Caspase 12, Fas, IL-15 und dem TNF-Rezeptor-assozierten Ligand (TRAIL) beobachtet. Poly(I:C) wird als eines der am besten geeigneten Produkte zur Generierung von stabilen ausgereiften dendritischen Zellen (DC) empfohlen. Diese ausgereiften DC können nach der Injektion aufgrund ihrer Fähigkeit, nach der CD40-Ligation bioaktives IL-12 zu sekretieren, in vivo effektive Immunreaktionen hervorrufen. Poly (I:C) wurde als potentes Adjuvans zur Verbesserung der spezifischen antitumoralen Immunreaktionen auf einen Peptid-Impfstoff verwendet.
Preparation Note
Das Produkt benötigt Ionenstärke, um die Doppelstrangstruktur aufrecht zu erhalten. Um eine Denaturierung zu vermeiden, sollte es in Lösungen mit physiologischer Salzkonzentration (z. B. Kochsalzlösung) rekonstituiert werden. Bei der Rekonstitution ist unter Umständen ein Erhitzen (50 °C) und erneutes Abkühlen erforderlich, um ein Re-Annealing zu erzielen.
