General description

Leptomycin B ist ein antimykotisches, antibiotisches Anti-Tumor-Zytotoxin, das die CRM1-abhängige, NES-abhängige nukleozytoplasmische Translokation hemmt (nuklearer Export-Inhibitor). Zu den NES-enthaltenden Proteinen gehören HIV-1 REV, Aktin, c-Abl, Cyclin-B1, MDM2/p53, I?B, MPF, PKA und MEK.

Biochem/physiol Actions

Leptomycin B ist eine ungesättigte Fettsäure mit verzweigter Kette und ein wichtiges Werkzeug bei der Untersuchung von nuklearem Export. Leptomycin B ist ein spezifischer Inhibitor von Proteinen, die ein nukleares Exportsignal enthalten. Leptomycin B hemmt die nukleozytoplasmische Translokation von Molekülen, wie z. B. des HIV-1 Rev-Proteins, und den Rev-abhängigen Export von mRNA. Die Zugabe von sehr geringen Mengen an Fibroblasten verursacht die Akkumulation von MEK im Nukleus. Weitere Proteine, die von Leptomycin B beeinflusst werden, sind Aktin, c-Abl, Cyclin-B1, MDM2/p53, IκB, MPF und PKA. Der vermutete Hemm-Mechanismus beinhaltet die direkte Bindung von Leptomycin B an CRM1, was die Bindung von CRM1 an das nukleare Exportsignal enthaltende Proteine mittels der Interaktion mit Zysteinresten in einer von CRM1 konservierten Region blockiert.
