Application

Lyticase aus Arthrobacter luteus wird zum Lysen der Pilzzellwand für die DNA-Isolierung eingesetzt.
Lyticase von Arthrobacter luteus wird wie folgt verwendet:
  • zur Herstellung von Sphäroplasten
  • als Komponente der Aufschlusslösung, zur Inkubation von Hefezellen zum Aufschluss der Zellwand
  • im Rahmen der enzymatischen Hydrolyse des Myzel-Präzipitats zur Herstellung von Protoplasten

Biochem/physiol Actions

Hefezellen sind schwer zu zerstören, da ihre Zellwand Kapseln oder resistente Sporen bilden kann. DNA kann durch Lyseenzyme wie Lyticase aus Hefe extrahiert werden, um die partielle Sphäroplastenbildung einzuleiten. Sphäroplasten werden anschließend lysiert, um DNA freizusetzen. Lyticase wird für den Aufschluss der Zellwand von Hefe und zum Erzeugen von Sphäroplasten aus Pilzen für die Transformation bevorzugt. Sie enthält β-(1→3)-Glucanlaminaripentaohydrolase und β-(1→3)-Glucanase-, Protease- und Mannanaseaktivitäten. Lyticase wird für Hefezellen wie Candida, Debaryomyces, Saccharomyces, Saccharomycopsis, Saccharomycodes, Eremothecium und Schwanniomyces-Spezies eingesetzt.
Das Enzym Lyticase wird häufig in der Pilzforschung eingesetzt, insbesondere zur Spezies-Identifizierung mittels PCR-basierter (Polymerase-Kettenreaktion) Verfahren. Es kann β-(1→3)- und β-(1→4)-Bindungen zwischen Glucoseeinheiten aufbrechen. Lysozym dient als Indikator für eine Makrophagen-vermittelte Wirtsreaktion, ist mit dem Leukozytentod korreliert und weist eine hohe Fluktuationsrate auf. Erhöhte Serumkonzentrationen von Lysozym werden bei verschiedenen chronischen Entzündungen, entzündlichen Darmerkrankungen, hämatologischen Erkrankungen und Nierenerkrankungen beobachtet. Das C-Typ-Lysozym aus Hühnereiweiß wird häufig als Modell zur Untersuchung der Proteinstruktur und -funktion verwendet. Muramidase zeigt in erster Linie eine bakterizide Wirkung gegen grampositive Bakterien. Die in der Mitteilung beschriebene Methode zur Bestimmung molekularer Massen mittels Gelfiltrationschromatographie ist eine modifizierte Version der bereits veröffentlichten Verfahren. Die in diesem Kit enthaltenen Proteinstandards sind möglicherweise mit anderen chromatographischen Systemen wie der Hochleistungs-Flüssigkeitschromatographie (HPLC) kompatibel. Bestimmte Puffersysteme können sich jedoch auf die Elutionsvolumina von Albumin und Carboanhydrase auswirken. Die Proteine in diesem Kit haben einen molekularen Massenbereich von 29 kDa bis 699 kDa.
Lyticase hydrolysiert Poly-β(1→3)-Glucose wie das Zellwand-Glucan der Hefe.

  1. Is this lyticase lyophilised powder(4.1 mg) (L2524) soluble in water. Because upon adding sterile nuclease free water(1 ml), i still can see some remains floating. what can i do? and what will be stability?

    1. This product can be reconstituted in cold deionized water to about 500 units per mL. Activity may decline quickly; use a freshly prepared solution. Do not store stock solutions. A 10× stock is described (1 µg/µL) with lyticase, buffer pH 7.5, NaCl, glycerol, and water stored at –20 °C, but stability in this product at –20 °C has not been validated. If particulates remain after reconstitution, mix thoroughly. Prepare a fresh solution before use.

  3. How many milligrams is 10,000 units of Product L2524 Lyticase from Arthrobacter luteus?

    1. The milligram quantity of this product will vary from lot to lot. The vial content is dependent upon the protein content and enzymatic activity. The minimum protein content will be 20% and the minimum activity will be 2000 units per milligram of protein. As an example, a batch that reports a protein content of 20% and an activity of 5000 units per milligram protein will have a vial content of 2 mg for the 10,000 unit package size.     1. There have been no tests conducted for product L2524 application on the fungus Aspergillus fumigatus. However, according to a citation in Vet Pathol, 2U/100 ul of fungal extraction has shown good results. The citation is titled "Panfungal Polymerase Chain Reaction for Identification of Fungal Pathogens in Formalin-Fixed Animal Tissues" and was published in July 2017. The DOI is 10.1177/0300985817698207, and the PMID is 28346123

  8. I see it is suggested to reconstitute Lyticase fresh. However, if that is not possible, can you store reconstituted Lyticase, L2524, at -20C? If so, for how long?

    1. The solution stability of this product has not been determined. However, according to Cold Spring Harbor Protocols, lyticase may be stored up to 6 months at 4 deg. C when prepared in 10 mM Tris-CL containing 20% glycerol. This has not been validated. See the link below to this citation:

  9. How can I store Product L2524, Lyticase, once reconstituted?

    1. When lyticase is reconstituted and stored, it may be stable for no more than 4-6 hours before serious loss of activity is observed. It is suggested that fresh solutions be prepared for use.
