Sodium channel protein type 9 subunit alpha (UniProt: Q15858; also known as Neuroendocrine sodium channel, hNE-Na, Peripheral sodium channel 1, PN1, Sodium channel protein type IX subunit alpha, Voltage-gated sodium channel subunit alpha Nav1.7, SCN9A) is encoded by the SCN9A (also known as NENA) gene (Gene ID: 6335) in human. Sodium channels are multi-pass membrane glycoproteins that mediates the voltage-dependent sodium ion permeability of excitable membranes. Assuming opened or closed conformations in response to the voltage difference across the membrane sodium-selective channel allow sodium ions to pass in accordance with their electrochemical gradient. Nine different sodium ion channels have been described and they have been classified based on their sensitivity to tetrodotoxin. Mammalian sodium channels are heterotrimers that are composed of a large central pore-forming glycosylated alpha subunit and 2 smaller auxiliary beta subunits. Quality Control Testing

Evaluated by Western Blotting in Rat brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected pan-Na+ Channel in Rat brain tissue lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected pan-Na+ Channel in human and mouse brain tissue lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected pan-Na+ Channel in E18 rat cortical cells.

Affinity Binding Assay: A representative lot of this antibody bound Sodium Channel-Pan peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Biochem/physiol Actions

Clone 4B8 is a ZooMAb^® rabbit recombinant monoclonal antibody that detects various sodium channels. It targets an epitope within 18 amino acids from the 10th cytoplasmic domain in the C-terminal half. It reacts with all known Sodium channel (SCN) alpha subunits.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
