The 395B kit is a kit formulated for multiple uses. Typically, the reagent is only replaced after the staining performance drops off. For kit 395B, calculating an exact range of slides is not possible. The kit is provided with 4 each of the 50 ml bottles of Schiffs reagent and 2 each for the 100 ml bottles for Periodic Acid. Each 50 ml aliquot is probably sufficient for 5-10 uses. The exact number will depend on several factors. If the slides are all to be stained at one time, a larger number of slides could be stained. If the slides are stained 2 or 3 at a time over a large number of weeks, fewer slides could be stained. Again, the solutions are to be replaced as needed and not necessarily after a set number of slides have been stained.

Application
Bei Behandlung mit Periodsäure werden Glycole zu Aldehyden oxidiert. Nach der Reaktion mit Schiffs Reagenz (einer Mischung von Pararosanilin und Natriumdisulfit) wird ein Pararosanilin-Addukt freigesetzt, das die glycolhaltigen Zellelemente pink bis rot färbt. Zellelemente, die mit der PAS-Reaktion nachgewiesen werden können, sind unter anderem Glycogen, Pilzzellwände, Basalmembranen, bestimmte Sulfomucine und Sialomucine in Epithelien, neutrale Mukosubstanzen, kolloidales Material der Schilddrüse und der Pars intermedia der Hypophyse.
