General description

28 kDa

Chromatin assembly factor-1 (CAF-1) ist ein Proteinkomplex mit mehreren Untereinheiten, der drei als p150, p60 und p48 bekannte Polypeptiduntereinheiten umfasst. CAF-1 ist ein Assemblierungsfaktor, der neu synthetisierte und acetylierte Histone H3/H4 bei der DNA-Replikation in entstehendes Chromatin einlagert.

Heterochromatin wurde als dicht gewickeltes Chromatin charakterisiert, das allgemein in der späten S-Phase repliziert eine geringe Gendichte aufweist und große Blöcke sich widerholender DNA enthält, die für DNA-Modifikationsreagenzien relativ unzugänglich is. In der späten S-Phase assoziiert p150 direkt mit Heterochromatin-assoziierten Proteinen 1 (HP1α, HP1β und HP1γ). Wenn Zellen sich auf die Mitose vorbereiten, dissoziieren CAF-1 p150 und einige HP1 schrittweise von Heterochromatin, was mit der Phosphorylierung von Histon H3 übereinstimmt. Die HP1-Proteine re-assoziieren am Ende der Mitose wieder mit dem Chromatin, wenn das Histon H3 dephosphoryliert wird.
