Beschreibung

General description

Der CD40-Ligand (CD40L/TRAP) ist ein Membranprotein vom Typ II, das für die Helferfunktion von T-Zellen an ruhenden B-Zellen verantwortlich ist. GF101 ist ein 16,3-kDa-Protein, das 149 Aminosäurenreste enthält und die rezeptorbindende TNF-ähnliche Domäne von CD40L umfasst.

Sterilfiltriert und lyophilisiert aus 10 mM Natriumphosphat, pH-Wert 7,5.

Preparation Note

Das lyophilisierte Protein ist bei Raumtemperatur ein paar Wochen stabil, sollte idealerweise aber bei -20 ºC aufbewahrt werden. In Wasser auf 0,1 bis 1,0 mg/ml rekonstituieren. Diese Lösung kann in anderen wässrigen Puffern verdünnt werden und bei 2–8 ºC eine Woche lang oder bei -20 ºC zur späteren Verwendung gelagert werden. Rekonstituiertes sCD40L/TRAP ist bei -20 ºC in unverdünnten Aliquoten bis zu 6 Monate haltbar. Gefrier-/Auftauzyklen haben eine signifikante Aktivitätsminderung zur Folge.

Analysis Note

Spezifische Aktivität: GF101 ist biologisch voll aktiv, was durch seine Fähigkeit ermittelt wurde, die Proliferation von HEK-BlueTMCD40-Reporterzellen zu stimulieren.
