Beschreibung

General description

Eine zellpermeable Tyrphostinverbindung, die als teilselektiver Deubiquitinase(DUB)-Inhibitor wirkt und den JAK-Stat-Signalweg unterdrückt. Es wird berichtet, dass sie eine Akkumulation von polyubiquitiniertem p53, JAK2 und Bcr-Abl verursacht sowie die c-Myc- und MCL-1-Proteinspiegel in verschiedenen Tumorzellen reduziert und die Aktivitäten von Stat3 und NF-κB herunterreguliert (IC₅₀<1 µM in MCL-Zellen), was zur Apoptose der Tumorzellen führt (IC₅₀ ~1,2 µM in Z138-Zellen). Es wird vorgeschlagen, das Cystein im aktiven Zentrum von DUB zu modifizieren und die Aktivitäten von USP5, UCH-L1, USP9x, USP14 und UCH37 zu hemmen (≥ 80%ige Hemmung bei 5 µM), ohne das 20S-Proteasom zu beeinflussen. Berichten zufolge weist sie stärkere antiproliferative und antitumorale Eigenschaften als der STAT3-Inhibitor III, WP1066 (Best.-Nr. 573097), in Xenograft-Mausmodellen von CML und Melanomen auf.
