Beschreibung

General description

Ein chromogenes β-Lactamase-Substrat, bei dem eine ausgeprägte Farbänderung von gelb (λ_max = 390 nm bei pH 7,0) nach rot (λ_max = 486 nm bei pH 7,0) auftritt, wenn die Amidbindung im β-Lactamring durch β-Lactamase hydrolysiert wird. Nitrocefin hydrolyseempfindlich gegenüber allen bekannten Lactamasen, die von grampositiven und gramnegativen Bakterien erzeugt werden. Auch hilfreich für den Nachweis von β-Lactamasemustern in bakteriellen Zellextrakten mittels isoelektrischer Fokussierung. Wurde in kompetitiven Inhibitionsstudien in Entwicklungsarbeiten zu β-Lactamase-resistenten Antibiotika verwendet.

Preparation Note

Herstellung einer Nitrocefin-Lösung (500 µg/mL)

• 1 mg Nitrocefin in 100 µL Dimethylsulfoxid (DMSO) auflösen und vortexen.

• 1,9 mL Phosphatpuffer (100 mM, pH 7) für ein Gesamtvolumen von 2 mL zugeben.

• Dies ergibt eine Nitrocefin-Arbeitslösung von 500 µg/mL (~ 1 mM), die für die meisten Anwendungen ausreicht.

• Nitrocefin ist insbesondere in Lösung sehr lichtempfindlich.

Nach der Rekonstitution in Aliquote aufteilen und einfrieren (-20 °C). Die Stammlösung kann bei -20 °C bis zu 2 Wochen aufbewahrt werden.
