Beschreibung

General description

Natives Calpain-1 aus humanen Erythrozyten. Ca²⁺-abhängige Cysteinproteinase mit geringem Ca²⁺-Bedarf (mittlere effektive Konzentration = 2 µmol/l). Ist an der ATP-Freisetzungsreaktion von mit Thrombin stimulierten Blutplättchen beteiligt.

Physical form

In 20 mmol/l Imidazol, 5 mmol/l β-Mercaptoethanol, 1 mmol/l EDTA, 1 mmol/l EGTA, 30 % Glycerin, pH-Wert 6,8.

Preparation Note

Aus Plasma hergestellt, das in zertifizierten Tests für HBsAg und Antikörper gegen HIV und HCV als negativ befunden wurde.

Nach dem erstmaligen Auftauen in Aliquote aufteilen und einfrieren (-70 °C).

Eine Einheit ist als die Menge Enzym definiert, die bei 25 °C unter Verwendung des Calpain-Aktivitätsassay-Kits, fluorogen (Katalognr. QIA120) in 1 min 1 pmol Suc-LLVY-AMC hydrolysiert. Hinweis: 1 caseinolytische Einheit = 1 fluorogene Einheit.
