General description

Macrophages and granulocytes express Signal regulatory protein (SIRP), which is a transmembrane glycoprotein involved in the down regulation of tyrosine kinase receptor signal transduction induced by cell adhesion, growth factors or insulin. They are immunoglobulin-like cell surface receptor for CD47. By CD47 binding there is a negative regulation of phagocytosis, mast cell activation and dendritic cell activation that prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells.

Specificity

Expected to cross-react with rat and mouse.
MAB1407P recognizes rat SIRP (Signal regulatory protein), which is expressed selectively by myeloid cells and by neurons. This monoclonal binds to a different epitope on SIRP than ED9. It does not block the interaction of CD172a and CD47.
Labels most macrophages including resident peritoneal, alveolar and activated macrophages. Recognition of other macrophages is limited to only occasional kupffer cells. In addition it labels granulocytes and a population of dendritic cells. It also labels brain extensively.

Immunogen

Rat peritoneal macrophages.

Application

Immunohistochemistry:
Frozen sections and Formalin fixed, paraffin embedded tissue sections (requires trypsin or pronase pre-treatment).

Western Blotting:
A previous lot of this antibody was used in western blot.

Flow cytometry:
1:100-1:200 of a previous lot was used. (Use 10 μL of working dilution for 10^6 cells in 100 μL).

Optimal working dilutions must be determined by the end user.
