General description

Rotenone belongs to the retinoid family. It is naturally present in Leguminosa plants and is considered to be cytotoxic. It is commonly used as an insecticide and a pesticide.[1]

Application

Rotenone has been used:
  • to study its effect on cell proliferation[2]
  • to detect the production of cellular reactive oxygen species[3] in isolated rat forebrain mitochondria[4]
  • to measure mitochondrial enzymatic activity[5]

Biochem/physiol Actions

Rotenone is an inhibitor of mitochondrial electron transport at nicotinamide adenine dinucleotide (NADH):ubiquinone oxidoreductase. It is readily absorbed through the exoskeletons of arthropods, but poorly absorbed cutaneously or from the gastrointestinal tract of mammals. Rotenone acts as a neurotoxic agent which can produce Parkinson-like condition to serve as an animal model for the study of etiology and interventions. The primary mechanism of action of rotenone is complex I inhibition, followed by the generation of oxidative stress and oxidative damage. Because rotenone is lipophilic, it readily penetrates the brain, where it attaches to and inhibits mitochondrial complex I of electron chain transport (ETC).

Preparation Note

This product is soluble in DMSO at 0.5 mg/ml. Other sources suggest it can be dissolved in DMSO up to 100 mM, however this has not been verified.

  1. For how long can the product maintain stability in powder form at 4 °C or -80 °C?

    1. The recommended storage temperature for this material is ambient or room temperature. Studies have not been conducted to evaluate the stability or shelf life impact of the product when stored at other temperatures.   2. Can aliquots of rotenone resuspended in DMSO be stored at -20C?

    1. The solution stability has not been determined. However, a similar product, 557368, notes that "following reconstitution, aliquot, flush with nitrogen, and store at -70°C. Stock solutions are stable for up to 1 month at -70°C".

  3. What is the solubility in other compounds ?

    1. This product is tested for solubility at 50 mg/mL in chloroform. Rotenone is poorly soluble in water at approximately 0.25-0.50 uM. Other options include Ethanol and DMSO.
      Ethanol: 5 mg/mL (higher concentrations may be achieved with heat and sonication)
      DMSO: 50 mg/mL
