General description

Alternative-splicing factor 1 (ASF-1), also known as SRp30a or SF2, is a member of the highly conserved family of SR proteins, a group of serine/arginine-rich domain-containing proteins that play a critical role in regulated and constitutive splicing of precursor mRNA. The SR family of proteins is characterized as having one to two RNA recognition motifs located in the n-terminal sequence, and an RS domain within the C-terminus that is rich in serine/arginine dipeptide repeats. ASF-1 has an inhibitory role in exon skipping, in order to maintain splicing accuracy and to monitor alternative splicing. Studies have shown that ASF-1 plays a role in the regulation of caspase-9 alternative splicing as well as being a critical component for ceramide to function as a mediator of alternative splicing for the same caspase.

Specificity

This antibody recognizes the N-terminus of ASF-1 (SF2).

Immunogen

Epitope: N-terminus
MBP-tagged recombinant protein corresponding to the N-terminus of human ASF-1 (SF2).

Application

Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected ASF-1 (SF2) in human prostate carcinoma cells and human testis tissue.
Western Blotting Analysis: A representative lot was used by an independent laboratory in western blotting (Sinha, R., et al. (2010). Mol. Cell. Biol. doi:10.1128/MCB.01270-09).
Immunoprecipitation Analysis: A representative lot immunoprecipitated ASF-1 (SF2) in HeLa nuclear extracts (Jablonski, J. A., et al. (2010). Nucleic Acids Research. 38(4):1249-1260).
