Cdk-activating kinase (CAK) is a trimeric complex composed of cyclin-dependent kinase 7 (Cdk7), cyclin H, and RING finger protein Methionine Adenosyltransferase 1 (MAT1). Cyclin H activates Cdk7, while MAT1 plays a role in influencing the substrate specificity of the complex. [1]

Acciones bioquímicas o fisiológicas

The Cdk7/cyclin H/Mat1 (CAK complex) plays a role in both cell cycle regulation and transcription. Within the complex, Cdk7 functions as the Cdk-activating kinase for cell cycle and as a part of the general transcription factor, Transcription Factor II H (TFIIH), for transcription process. [1] Additionally, it also regulates gene expression by phosphorylating transcription factors such as estrogen receptor-α (ER), p53, retinoid receptors, androgen receptor, and estrogen receptor.[2] Elevated expression levels of Cdk7, cyclin H, and MAT1 have been observed in breast cancer patients.[2] Inhibition of cdk7 is associated with the development of colorectal cancer in humans.[3]
