Application

Superfibronectin from human plasma dramatically enhances the adhesive properties of fibronectin and suppresses cell migration.[1][2] Improved results have been shown CHO cells, IMR-90 fibroblasts, T47D breast carcinoma cells, and UCLA-P lung carcinoma cells. Additional applications include epithelial cells, mesenchymal cells, neuronal cells, fibroblasts, neural crest cells, and endothelial cells. It is recommended for use as a cell culture substratum at 1 μg/cm2.

Components

Superfibronectin is formulated with recombinant, human fibronectin fragment III-C expressed in E. coli and fibronectin from human plasma. The recombinant fibronectin includes a new region, type III, cloned from human placenta cDNA, that is newly reported to participate in matrix formation by inducing spontaneous disulfide crosslinking of fibronectin molecules into multimers. The product resembles the in vivo matrix form of fibronectin.

Caution

Superfibronectin should be stored at 2-8°C and diluted immediately prior to use. Do not freeze this product.

Preparation Note

As supplied, this product is a 2 mg protein/mL solution in 0.05 M Tris buffered saline. Before coating a 96 well plate, it is recommended to dilute t he product with sterile PBS to a concentration of 5 μg/mL and add 100 μl/well. These plates should then be incubated for 2 hours at 37°C and washed twice with 100 μl of PBS. The source material has been tested for HBsAg and HIV antibodies.
