General description

The integrin family of cell adhesion receptors consists of at least 19 membrane-associated heterodimers, composed of an alpha and beta subunit that associate in a non-covalent manner. To date, 16 different alpha-subunits and 8 different beta-subunits have been identified. The structure and functional diversity of the integrin family are based upon the pairing abilities of the individual alpha and beta subunits. Key to these molecular interactions between the integrin receptors and their respective ligands is the recognition of the Arg-Gly-Asp (RGD) sequence, known to be present in the extracellular matrix components fibronectin, vitronectin, collagen, fibrinogen, and von Willebrand factor (Cheresh, 1991). The involvement of integrins in vascular proliferation, adhesion, and wound repair has been well documented. Integrin alphaV is a type I membrane glycoptroein with no I (inserted) domain. The alphaV protein is post-translationally cleaved into a heavy and light chain, which is expressed on the cell surface as a disulfide linked polypeptide.

The integrin alphavbeta6 is expressed on a variety of epithelial cells during dynamic processes including organogenesis, tissue injury and malignant transformation.

Specificity

Monoclonal antibody MAB2077Z, raised by immunizing beta6 knockout mice with wild type murine keratinocytes, recognizes human and murine alphaVbeta6 by flow cytometry and immunoprecipitation and has blocking activity against murine and human alphaVbeta6. 100% inhibition of alphaVbeta6-mediated cell adhesion and cell migration are achieved at a concentration of <10 μg/mL. Because the antibody was raised in knockout mice, it is likely to recognize the receptor in other species, however it has been evaluated only in human and mouse.The antibody is specific for alphaVbeta6 and does not recognize aV or other aV integrins.

Immunogen

Immunization of beta 6 knockout mice with wild type murine keratinocytes.

Application

Anti-Integrin αVβ6 Antibody, clone 10D5, azide free is an antibody against Integrin αVβ6 for use in FC & IP.
Flow cytometry: Effective for mouse and human.

Immunoprecipitation: Effective; however because this antibody is an IgG2a, protein G or rabbit anti-mouse IgG is helpful for capture

Function blocking: 100% inhibition of alphaVbeta6-mediated cell adhesion and cell migration are achieved at a concentration of 100 μg/mL.

Not effective for western blot or immunohistochemistry.

Optimal working dilutions must be determined by end user.
