General description

Y. enterocolitica Millichrome plus Supplement is a selective media supplement for the detection of Yersinia enterocolitica. The distinguishing feature of these media is the inclusion of a chromogenic substrate mix that produces visible color changes upon enzymatic activity of Yersinia enterocolitica.Millichrome chromogenic media is a specialized type of culture medium used to detect and isolate specific groups of bacteria based on their ability to produce enzymes with chromogenic substrates. The package contains 1 vials sufficient for 1 liter of final media. The prepared supplement is added to 1 liter of melted Y. enterocolitica Millichrome plus Agar base cooled at 45-50 °C +/- 2 °C.

Application

Y. enterocolitica Millichrome plus Supplement is a selective chromogenic media supplement intended for use in the qualitative detection, differentiation, and presumptive identification of Yersinia enterocolitica from various samples.

Components

Composition per vial:
(1 vial is sufficient for 5 L of final media)
Selective mix 0.5g

Preparation Note

  • Prepare a stock solution of the supplement: Add 100 mg to 1 mL of purified water.
  • Swirl well until complete dissolution. Filter sterilize at 0.45 μm.
  • Warning 2: This supplement stock solution should be used immediately after Preparation, or can be stored at -20 °C and used within 15 days.
  • Add 1 mL of the prepared supplement solution to 1 L of the prepared Y. enterocolitica Millichrome plus Agar base cooled at 45-50 °C +/- 2 °C.
  • Swirl gently to homogenize.
  • Pour into sterile Petri dishes.

Analysis Note

The Pathogenic Y. enterocolitica is detected by a characteristic mauve color after only 36-48 h of incubation, while the non-pathogenic Y. enterocolitica and background flora (Citrobacter, Enterobacter, Aeromonas, etc.) form metallic blue colonies or limited growth in metallic blue color after incubation at the same temperature
