Gene Information

human ... MKNK1(8569)

General description

MNK1 is a serine-threonine kinase that is activated by ERK or p38 MAPK signaling cascades and modulates translation through eIF4E phosphorylation. Human MNK1 has two C-terminal splice variants, namely, MNK1a and MNK1b. While MNK1a is expressed in the cytoplasm, MNK1b is mainly localized at the nucleus . Anti-MNK1 antibody is specific for human Mnk1 (40 kDa). In immunoblotting, staining of the Mnk1 band is specifically inhibited by the immunizing peptide.

Immunogen

synthetic peptide corresponding to amino acids 12-27 of human MNK1, conjugated to KLH.

Application

Anti-MNK1 antibody is suitable for use in western blot (1-2 μg/mL using lysates of HEK-293T cells transfected with human Mnk1).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

The human MAP kinase-interacting kinases (or MAP kinase signal-integrating kinases), Mnks, comprise a group of four proteins derived from two genes (Gene symbols: MKNK1 and MKNK2) by alternative splicing. Mnk1a/b differ at their C-termini, as do Mnk2a/2b: in each case

Mnk1 and Mnk2 are protein kinases that are directly phosphorylated and activated by extracellular signal-regulated kinase (ERK) or p38 mitogen-activated protein (MAP) kinases and implicated in the regulation of protein synthesis through their phosphorylation of eukaryotic translation initiation factor 4E

In this paper, we report the identification and molecular characterization of a splice variant of human Mnk1 which has been named as Mnk1b. Human Mnk1b mRNA is homologous to human Mnk1 mRNA but lacking a region corresponding to exon 19

We have developed a novel expression screening method for identifying protein kinase substrates. In this method, a lambda phage cDNA expression library is screened by in situ, solid-phase phosphorylation using purified protein kinase and [gamma-32P]ATP. Screening a HeLa cDNA library
