General description

Lidocaine is a local anesthetic and an effective antiarrhythmia agent. It occurs as solutions of crystalline colorless solid for injections, that has a short half-life within plasma.

Application

Lidocaine may be used for drug testing and for voltage clamp experiments.

Biochem/physiol Actions

Na+ channel blocker; class IB antiarrhythmic that is rapidly absorbed after parenteral administration.
Na+ channel blocker; class IB antiarrhythmic.
Lidocaine, a neuroprotective agent, mediates blockage of sodium (Na+) channels. It prevents the influx of sodium ions leading to depolarization of sodium channels and metabolically inactivated in liver. Lidocaine has positive effect on cerebral ischemia. It is also used in the treatment of ventricular tachycardia. Lidocaine is an efficient anesthetic agent in ophthalmic surgeries as it is highly potent and has good tissue penetrability.

Questions

1–4 of 4 Questions
  1. How does the storage temperature relate to shipping conditions?

    1 answer
    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

  3. Is Product L7757, Lidocaine free base, suitable for use as a reference standard?

    1 answer
    1. No. We would recommend Product No. L1026 (Lidocaine, Sigma References Standard). The current lot is current with USP 31, 2nd Supplement, and is traceable to USP Reference Standard lot MOF220.

  4. How do I dissolve lidocaine free base (Product No. L7757)?

    1 answer
    1. The product is soluble in alcohol, ether, benzene, chloroform and oils, according to the chemicals encyclopedia published by the Royal Society of Chemistry , 14th ed. online  (2006).  Lidocaine hydrochloride monohydrate (Product No. L5647) is soluble in water.  Since the lidocaine hydrochloride is so much more soluble than lidocaine free base, lowering the pH to that of the hydrochloride (pH 4.0-5.5) or to that of injectable forms (pH 5.0-7.0) should greatly enhance the solubility of L7757.
