General description

Endothelin consists of a family of potent vasoconstrictor peptides, which include four structurally related isoforms, ET-1, ET-2, ET-3 and vasointestinal contractor (VIC, β-ET).

Specificity

Reacts with endothelin-1 (ET-1), (human, porcine), endothelin-2 (ET-2), (human), and endothelin-3 (ET-3), (human, rat). Cross-reactivity is seen with sarafotoxin S6c, but not with big-endothelin 38 (human), big-endothelin 39 (rat) or atrial natriuretic peptide (human). There is minimal cross-reaction with angiotensin I (human).

Application

Anti-Endothelin antibody produced in rabbit is suitable for immunodetection of endothelins (ET-1, ET-2, and ET-3) in various immunoassays including RIA (titer of 1:30,000) and ELISA (titer of 1:10,000). Antibodies that react specifically with endothelins (ET-1, ET-2, and ET-3) may be used to detect endothelins in tissue extracts and biological fluids as well as study differential tissue expression, and intracellular localization of the endothelin isoforms in the periphery and central nervous system.

Biochem/physiol Actions

Endothelins (ET) show potent constrictor activity in vascular and non-vascular smooth muscle. ETs can mediate biological effects in cells and tissues, and have been shown to bind to an ET receptor in the lung, kidney, heart, and liver. The biological actions of ETs are mediated by activation of phospholipase C through specific G protein-coupled receptors.
