General description

We are committed to bringing you Greener Alternative Products, which adhere to one or more of The 12 Principles of Greener Chemistry. Laccase is an enzyme, produced by ericoid mycorrhiza and ectomycorrhiza fungi. It belongs to the group of polyphenol oxidases. Laccase is also present in plants and bacteria.[1]

Application

Laccase from Trametes versicolor has been used:
  • to assess the use of four laccase-producing strains in waste water treatment[1]
  • in laccase assay[2]
  • in screening the lignols

Biochem/physiol Actions

Laccase catalyzes the oxidation of phenol containing compounds, including lignin, through the reduction of oxygen to water. The presence of mediators will allow the oxidation of non-phenlic compounds as well. The primary function of laccase is to degrade lignin in fungi.
Some of the enzymatic actions of laccase are associated with sporulation, detoxification, morphogenesis, melanin polymerization and it offers protection to spore coat. Laccase can catalyse a number of substrates including medicinal drugs and halogenated pesticides. It utilizes oxygen for its catalysis. For these reasons, it might be useful in the biological degradation of micropollutants in wastewater treatment.[1]

  1. I dissolved 1 mg this product in 1 ml buffer (expected 1 mg/ml), but Bradford assay showed 0.01 mg/ml. Why is there such a big difference?

With product 38429, the Enzymatic Activity specification per each lot is > or = to 0.5 (in U/mg).  1 U Corresponds to the Amount of Enzyme which Converts 1 mol Catechol per Minute at ph5.0 and 25 deg C.  For each lot of product received, please consult the Certificate of Analysis to see how much is in each lot.  From there, please calculate the concentration desired for your application.

    1. The molecular weight of this product has not been determined. Literature sources indicate that fungal laccase has a molecular weight range of 60 - 70 kDa; however, this may be significantly influenced by glycosylation. Please see the link below to review a publication that may be helpful:

  5. What is the optimum temperature and optimum pH-value for this laccase? Is there an exact value or a range available?

    1. According to the procedure for the enzymatic assay of laccase, the conditions are set at T = 30 °C and pH = 6.5.

    1. The minimum activity specification for this product is 0.5 µ/mg. This value varies from lot to lot. A review of several recent lots reveals a general range of 0.5 µ/mg – 1.1 µ/mg,

      A 1-gram vial with an activity of 0.5 U/mg would contain 500 units. The same lot in the 10-gram vial would contain 5000 units.
      The purity of this product is not tested. The presence or absence of any other non-enzyme admixtures has not been determined and such values are unavailable.
