General description

Phosphatidylinositol 5-phosphate 4-kinase type-2 beta (EC 2.7.1.149; UniProt P78356; also known as 1-phosphatidylinositol 5-phosphate 4-kinase 2-beta, Diphosphoinositide kinase 2-beta, Phosphatidylinositol 5-phosphate 4-kinase type II beta, PI(5)P 4-kinase type II beta, PIP4KII-beta, PIP4K2B, PtdIns(5)P-4-kinase isoform 2-beta) is encoded by the PIP4K2B (also known as PIP5K2B, PIP5KIIB) gene (Gene ID 8396) in human. PIP4K2B catalyzes the conversion of 1-phosphatidyl-1D-myo-inositol 5-phosphate to 1-phosphatidyl-1D-myo-inositol 4,5-bisphosphate. It is highly expressed in brain, heart, pancreas, skeletal muscle and kidney, however, lower levels are detected in placenta, lung, and liver. Mice deficient in PIP4K2B are shown to be hypersensitive to insulin and exhibit reduced body weights compared to wild-types. Adult male mice lacking PIP4K2B have significantly less body fat, but female mice show increased sensitivity to insulin in the presence of normal adiposity. Mice lacking PIP4K2B exhibit enhanced in vivo insulin-induced activation of Akt in the skeletal muscle and liver. Ref.: Lamia, KA., et al. (2004). Mol. Cell 24, 5080-5087

Specificity

Epitope region is present in spliced isoform 1, but not in isoform 2 reported by UniProt (P78356).

GST-tagged recombinant fragment corresponding to a region within the C-terminal half of human PIP4K2B (PIP5K2B).

Target description

~48 kDa observed. 47.25/47.19 kDa (human/mouse) calculated (Met 1 removed). Uncharacterized bands may be observed in some lysate(s).
