General description

TRIM63, also known as E3 ubiquitin-protein ligase TRIM63, Iris RING finger protein, Muscle-specific RING finger protein 1, MuRF-1, MuRF1, RING finger protein 28, Striated muscle RING zinc finger protein, Tripartite motif-containing protein 63, and encoded by the gene name TRIM63/ IRF/ MURF1/ RNF28/ SMRZ, is a E3 ubiquitin ligase. TRIM63 mediates the ubiquitination and subsequent proteasomal degradation of CKM, GMEB1 and HIBADH. It also regulates the proteasomal degradation of muscle proteins under amino acid starvation, where muscle protein is catabolized to provide other organs with amino acids. TRIM63 inhibits de novo skeletal muscle protein synthesis under amino acid starvation, regulates proteasomal degradation of cardiac troponin I/TNNI3, and probably of other sarcomeric-associated proteins. TRIM63 may play a role in striated muscle atrophy and hypertrophy by regulating an anti-hypertrophic PKC-mediated signaling pathway. TRIM63 may regulate the organization of myofibrils through TTN in muscle cells. TRIM63 is expressed selectively in heart and skeletal muscle. TRIM63 is also expressed in the iris.

Immunogen

KLH-conjugated linear peptide corresponding to the C-terminus of Human TRIM63.

Application

Immunohistochemistry Analysis: A representative lot of this antibody at 20 µg/mL detected Trim63 protein in human cardio myocyte. Similar staining was observed in human skeletal myocyte tissue at 4 µg/mL dilution
This Anti-TRIM63 antibody is validated for use in WB, IH(P) for the detection of TRIM63.

Quality

Evaluated by Western Blotting on human skeletal muscle lysate
Western Blotting Analysis: 1 µg/mL dilution of this antibody detected Trim63 protein in 10 µg of human skeletal muscle lysate

Target description

~40 kDa observed. Uncharacterized bands may be detected at high antibody concentrations or longer exposures
