The core histones, H2A, H2B, H3, and H4, form a central octameric structure that binds approximately 147 bp of DNA to form a nucleosome. The H2AZ histone variant replaces conventional H2A in a subset of nucleosomes and plays an essential role in transcriptional regulation, autosilencing, gene silencing, and genome stability. The effects of histone variants and histone modifications on chromatin structure and gene expression can be deciphered by applications such as chromatin immunoprecipiation (ChIP), which requires high quality chromatin and essential tools, such as histone proteins and appropriate enzymes to assemble chromatin in vitro.
