General description

Sigma offers four sizes of gradient mixers. The small, 15 and 30 mL mixers are ideal for the preparation of centrifugation gradients, using sucrose or cesium chloride solutions. The larger, 50 and 100 mL mixers are mainly used for the preparation of linear polyacrylamide gel gradients. The 100 mL mixer is configured to be used in conjunction with the multicaster systems. All of the mixers are produced from a single acrylic block. They consist of two chambers, a reservoir and a mixing chamber, with an interconnecting valve. A second valve regulates the output flow from the mixing chamber. A magnetic stirring bar can be placed in the mixing chamber to maintain a constant gradient. All of the mixers have a flat base to enable them to be placed on a magnetic stirrer. They also have a support rod which can be used to fix the mixers to a stand for extra stability.

Although the starvation response of the model multicellular organism Caenorhabditis elegans is a subject of much research, there is no convenient phenotypic readout of caloric restriction that can be applicable to large numbers of worms. This paper describes the distribution

Questions

  1. Please provide the instructions for using the Sigma-Aldrich Gradient Mixer, size 50 mL, catalog number Z340413.

    1 answer
    1. The product manual for the Sigma-Aldrich Gradient Mixer with catalog number Z340413 is not available. However, the Gradient Mixer is made from a single acrylic block, has two chambers (a reservoir and a mixing chamber) with an interconnecting valve, and a second valve to regulate the output flow. A magnetic stirring bar is used to maintain a constant gradient, and the mixer has a flat base for placement on a magnetic stirrer. It is recommended to ensure that the volume of solution in each chamber is inversely proportional to the densities of each and that the volumes used in the chambers are carefully controlled to allow for the small volume of medium retained by the gradient maker and the outlet tubing. For further guidance on gradient preparation, it is suggested to consult a suitable textbook, such as "Methods of Cell Separation" by P.T. Sharpe, published by Elsevier.
