Application

14-3-3 Zeta, histidine-tagged human was used to study its role in regulating the von Willebrand factor (VWF) binding functions of GPIb-IX. Zeta isoform of 14-3-3 protein facilitated the formation of human tau fibrillar polymers.[1]

Biochem/physiol Actions

14-3-3 ζ functions in cell signaling, cell cycle control, and apoptotic death.
14-3-3 protein is an adapter protein that inhibits apoptosis as well as differentially regulates MAPK cascades.[2] It facilitates the regulation of several cellular processes like- cell-cycle control, apoptosis, stress response and malignant transformation.[3]
The 14-3-3 proteins are a multifunctional protein family comprising seven mammalian isoforms that interact with over two hundred different intracellular molecules including kinases, phosphatases, transcription factors, scaffold proteins, and DNA. 14-3-3 proteins are linked to many human diseases. 14-3-3 ζ functions in cell signaling, cell cycle control, and apoptotic death. 14-3-3 ζ interacts with β-catenin and facilitates its activation by AKT. 14-3-3 ζ plays an important role in regulating von Willebrand factor (VWF) binding the function of GPIb-IX suggesting a role in preventing or treating thrombosis. 14-3-3 ζ is involved in various types of cancer as a prognostic and therapeutic target and also involved in Alzheimer′s disease.
