This gene encodes a member of a family of basic helix-loop-helix transcriptional repressors. The protein product of this gene, which is activated downstream of the Notch pathway, regulates cell differentiation in multiple tissues. Hes family bHLH transcription factor 5 (HES5) is a regulator of Wnt family member 3A (Wnt-3A) protein. It is involved in stimulating the proliferation and differentiation of hepatocellular carcinoma cells. Methylation of the HES5 promoter has been studied to have a link with prostate cancer.
