Biochem/physiol Actions
General transcription factor for RNA Polymerase II.
The amino terminal region of TFIIB contains a zinc ribbon fold composed of antiparallel Β strands. Mutational studies show that this domain is required for recruitment of RNA Pol II for transcriptional initiation.
Unit Definition
One gel shift unit is the amount of TFIIB required to form 0.2 pmol of TFIIB/TFIID/DNA complex as detected by gel shift.
Physical form
Solution in 20 mM Tris-HCl, pH 7.4, 100 mM NaCl, 2 mM DTT, 1 mM EDTA, 0.1% Triton® X-100.

The function of the conserved zinc-binding domains in the related Pol II- and Pol III-specific factors TFIIB and Brf was investigated. Three-dimensional structure modeling and mutagenesis studies indicated that for both factors, the functional surface of the zinc ribbon fold
