immunohistochemistry: suitable using 4% formaldehyde, paraffin-embedded human and mouse tissue immunoprecipitation (IP): suitable western blot: 1-4 μg/mL using whole extracts of Cos-1 cells transfected with human TCF-4
Transcription factor 4 or T-Cell Factor-4 (TCF-4), also called as transcription factor 7 like 2 (TCF7L2), is mapped to human chromosome 18q21.1. The gene codes for a basic helix-loop-helix DNA binding protein, which belongs to TCF family and is implicated in transcriptional regulation. It is expressed predominantly in nervous system. TCF4 protein has a transcriptional activation domain 2 (AD2), a transcriptional activation domain (AD1) and a nuclear localization signal (NLS) in the N-terminal end and a C-terminal basic helix-loop-helix (bHLH) domain, involved in dimerization and deoxyribonucleic acid (DNA) binding.
Immunogen
His-tagged fusion protein corresponding to amino acids 31-331 of human TCF-4
Application
May be used for gel-shift assays
Biochem/physiol Actions
Transcription factor 4 (TCF-4) interacts functionally with β-catenin to mediate Wnt signaling pathway and transactivates downstream target genes implicated in cancer development. TCF-4 plays a vital role in development of the nervous system and vertebrate development. Aberrations or mutation of the gene leads to Pitt-Hopkins syndrome (PTHS), a rare developmental disorder characterized by severe motor and mental retardation, typical facial features and breathing abnormalities.
Physical form
Solution in 0.7 M tris-glycine, pH 7.4, containing 0.105 M sodium chloride, 30% glycerol and 0.035% sodium azide.

T cell factor 4 (TCF4) interacts with beta-catenin in the WNT signaling pathway and transactivates downstream target genes involved in cancer progression. To identify proteins that regulate TCF4-mediated biological responses, we performed a yeast two-hybrid screen to search for a

γ-Catenin, an important component of desmosomes, may also participate in Wnt signaling. Herein, we dissect the role of γ-catenin in liver by generating conditional γ-catenin knockout (KO) mice and assessing their phenotype after bile duct ligation (BDL) and diethylnitrosamine-induced chemical

Transcription factor TCF4 (alias ITF2, SEF2 or E2-2) is a broadly expressed basic helix-loop-helix (bHLH) protein that functions as a homo- or heterodimer. Missense, nonsense, frame-shift and splice-site mutations as well as translocations and large deletions encompassing TCF4 gene cause
A recent study has shown that phosphorylated c-Jun (p-c-Jun) interacts with TCF4 to form a complex that cooperatively enhances their transcriptional activity in the presence of beta-Catenin, and that their interaction is critical for mouse intestinal tumorigenesis. To determine the

Osteopontin (OPN) is a multifunctional protein implicated in mammary development, neoplastic change, and metastasis. OPN is a target gene for beta-catenin-T cell factor signaling, which is commonly disturbed during mammary oncogenesis, but the understanding of OPN regulation is incomplete. Data
