General description

The development and maintenance of central and peripheral nervous system is controlled by a family of Trk family of neurotrophin receptors. Neurotrophins have an important role in development of the nervous system and mediate the cellular effects by binding to the Trk receptors. The Trk family consists of 3 members, TrkA (NTRK1), Trk B (NTRK2) and TrkC (NTRK3). Neurotrophin-3 (NT3) is the primary ligand for TrkC receptor, but it can also bind and activate the other Trk members.Trk receptors reportedly play a major role in the development and behaviour of neuroblastomas and other type of cancers. TrkC is expressed in at least 25% of primary neuroblastomas. Trk members have been observed to be over-expressed in congenital fibrosarcoma, papillary thyroid carcinomas, pediatric carcinomas and leukemias. Aberrations in TrkC gene have been reported in secretory breast cancers and acute myelogenous leukemia
Anti-TrkC recognizes human TrkC. The antibody shows less than 1% cross-reactivity with human TrkB.

Immunogen

purified recombinant human TrkC extracellular domain, expressed in NSO cells.

Application

Anti-TrkC may be used to detect human TrkC by immunoblotting using a concentration of 0.1-0.2 μg/mL. Neutralization of receptor-ligand interaction may be possible at a working concentration of 1-3 μg/mL which blocks 50% of the ligand binding. Neutralization of TrkC has been demonstrated in three-dimensional cultures of rat Sertoli cells. A working concentration of 15 μg/mL may be used for detection of human TrkC in paraffin-embedded sections. For flow cytometry applications, antibody concentration of 3-10 μg/mL/106 is recommended.

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline with 5% trehalose.
