General description

TWEAK (TNF-related weak inducer of apoptosis) belongs to the TNF (tumor necrosis factor) family of ligands. TWEAK is expressed in a variety of tissues, including the adult heart, pancreas, skeletal muscle, small intestine, spleen and peripheral blood lymphocytes. It exists in two active forms: membrane-associated mTWEAK and soluble type sTWEAK. The TWEAK gene is mapped to human chromosome 17p13.1. The protein has an amino terminal hydrophobic region.
Recombinant human TWEAK is a soluble 17.0kDa polypeptide (154 amino acid residues) comprising the TNF homologous region of TWEAK and is generated by proteolytic processing of the full length membrane anchored TWEAK protein.

Biochem/physiol Actions

TWEAK (TNF-related weak inducer of apoptosis) binds to fibroblast growth factor-inducible 14 (Fn14). It is mainly involved in nuclear factor κB (NF-κB) pathway and is responsible for cellular proliferation, migration, cell death, differentiation, angiogenesis and inflammation. TWEAK is associated with various disorders, including atherosclerosis, heart failure, type 2 diabetes, ischemic stroke, multiple sclerosis and chronic kidney disease. This gene is upregulated after tissue injury and in tumors.

Lyophilized from 10mM Sodium Phosphate, pH 7.5 + 80 mM L-Arginine.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/mL. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
