Biochem/physiol Actions

RU-SKI 43 is a potent and selective inhibitor of Hedgehog acyltransferase (Hhat) with an IC50 of 850 nM. RU-SKI 43 is the first small molecule found that inhibits the activity of Hedgehog acyltransferase (Hhat) and inhibits palmitoylation of Shh by endogenous Hhat in vitro and in cells. Most inhibitors of hedgehog signaling act downstream, inhibiting Smo or Gli components of the Shh pathway. The upstream activity of RU-SKI 43 directly on palmitoylation of the Shh ligand is unique.
RU-SKI 43 is a potent and selective inhibitor of Hedgehog acyltransferase (Hhat).
