General description

Tumor protein p53, also called as guardian of the genome, is encoded by the gene mapped to human chromosome 17p13.1.[1] p53 is a sequence-specific transcription factor[2] containing transactivation domain, proline-rich domain, specific DNA-binding domain (DBD) and a tetramerization domain. In addition, it also contains a nonspecific DNA-binding domain that might be involved in DNA damage recognition.[3]

Biochem/physiol Actions

Tumor protein p53 prevents tumor progression through transcriptional regulation of targeted genes involved in cell cycle arrest, DNA repair, apoptosis and cell senescence.[2] Mutation in the gene leads to the development of various types of human cancers including liver fluke-associated cholangiocarcinoma (CCA).[2] p53 also cause Li-Fraumeni syndrome (LFS) in its mutated form.[1]
