General description

Monoclonal Anti-C10orf54 (mouse IgG1 isotype) is derived from the hybridoma OR.4 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide.C10orf54 (Chromosome 10 open reading frame 54) is also referred to as GI24, B7 member B7-H5, VISTA (V-domain Ig suppressor of T cell activation) and Dies1. It is a novel surface glycoprotein belonging to the B7 immunoglobulin superfamily. It consists of a single IgV domain with 3 additional cysteine residues and a cytoplasmic tail. It is expressed on mature myeloid APCs (antigen-presenting cells) including peritoneal macrophages, mature BMDC (bone marrow-derived dendritic cell), neutrophils and CD11c+ DCs, and to a lesser quantity in T cells and activated Treg cells.

Application

Phosphotyrosine antibody inhibitor has been used in flow-cytometry and immunohistochemistry.

Biochem/physiol Actions

Studies suggest that C10orf54 (Chromosome 10 open reading frame 54) functions as a co-inhibitory ligand and restricts the mechanism of T cell proliferation and cytokine production. It was also suggested that C10orf 54 acts as a positive regulator as well as a substrate of MT1-MMP (trans-membrane type 1-matrix metalloproteinase) during tumor growth of collagen matrix, which regulates invasive growth.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
