Description

General description

Villin is also known as villin-1 or VIL1 is a Ca²⁺ -regulated actin-binding protein. It belongs to the gelsolin family. Villin is localized in the apical cytoplasm and brush borders of epithelial cells of the small and large intestinal mucosa, intrahepatic bile ducts, kidney proximal tubules, gall bladder epithelium and pancreatic ducts. It is located on the human chromosome at 2q35.

Specificity

Anti-Villin-1 (internal) specifically recognizes human villin-1.

Immunogen

synthetic peptide corresponding to an internal sequence of human villin-1, conjugated to KLH. The corresponding sequence has high homology (81% identity) to mouse villin-1.

Biochem/physiol Actions

Villin functions in modulating the structure and assembly of actin filaments. It also plays a role in modulating cell migration by phosphorylating villin at Tyr-60, Tyr-81 and Tyr-256.Villin plays a role in microvillus assembly and acts as a marker for epithelial cell differentiation. Expression of villin is stimulated during intestinal metaplasia observed in Barrett′s esophagus and chronic atrophic gastritis. Expression of villin is increased during human adenocarcinomas, such as colorectal cancer.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
