General description

Zinc finger protein 746 (ZNF746) or PARIS (parkin interacting substrate) contains a kruppel-associated box (KRAB) at its N-terminal and a C2HC/C2H2 type zinc finger at its C-terminal. It belongs to the KRAB-zinc-finger protein (ZFPs) group of transcriptional repressors. ZNF746 is highly conserved among human, mouse and rat, and widely expressed in many tissues. ZNF746 gene is mapped to human chromosome 7q36.1.

Specificity

Anti-ZNF746(C-terminalregion) specifically recognizes human ZNF746.

Immunogen

synthetic peptide corresponding to a sequence at the C-terminal region of human ZNF746, conjugated to KLH. The corresponding sequence is identical in human ZNF746 isoform 2, and in mouse and rat ZNF746.

Application

Anti-ZNF746 (C-terminal region) antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Zinc finger protein 746 (ZNF746) is regulated by the E3 ubiquitin ligase, parkin. It represses the expression of the transcriptional coactivator, peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α). It also targets nuclear respiratory factor 1 (NRF-1). ZNF746 has been shown to be accumulated in Parkinson′s disease (PD) models and has been suggested to play a role in the progress of PD. Elevated expression of ZNF746 is implicated in the loss of dopamine (DA) neurons in the substantia nigra. This loss of DA neuron is reversed by either coexpression of parkin or PGC-1α. It may serve as a target to inhibit non-small cell lung cancer (NSCLC) invasion.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
