General description

Monoclonal Anti-OTUB1 (mouse IgG1 isotype) is derived from the hybridoma OTU14 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice. OTU domain, ubiquitin aldehyde binding 1 (OTUB1) is a cysteine protease. It exists in two isoforms obtained by alternative splicing. OTUB1 belongs to the ovarian-tumor-domain-containing proteases (OTU) family of deubiquitinating enzymes. It is located on the human chromosome at 11q13.1.

Specificity

Monoclonal Anti-OTUB1 recognizes human OTUB1.

Immunogen

synthetic peptide corresponding to a sequence at the N-terminus of human OTUB1, conjugated to KLH. The isotype is determined by ELISA using Mouse Monoclonal Antibody Isotyping Reagents (Sigma ISO-2).

Application

Monoclonal Anti-OTUB1 antibody produced in mouse may be used in immunoblotting and immunocytochemistry.

Biochem/physiol Actions

OTU domain, ubiquitin aldehyde binding 1 (OTUB1) is implicated in mediating lymphocyte antigen responsiveness by stabilizing lymphocyte specific E3 ligase gene related to anergy in lymphocytes (GRAIL) in CD4+ T lymphocytes. OTUB1 is also found in Lewy bodies of the brain and it might be involved in the pathogenesis of neurodegenerative disorders. Upregulation of OTUB1 in mesangial cells (MC) play a role in decreasing decorin levels, thus affecting glomerulonephritides. Overexpression of OTUB1 is associated with hyperactivation of mitogen-activated protein kinase (MAPK) signaling and promote tumorigenic transformation.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For extended storage, freeze at -20 °C in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
