General description

Proline-rich Akt substrate of 40 kilodalton (PRAS40), also known as AKT1 substrate 1, is encoded by the gene mapped to human chromosome 19q13.33.

Immunogen

synthetic phospho-peptide containing phosphorylated Thr246 of human PRAS40, conjugated to KLH. The corresponding sequence is identical in mouse and rat PRAS40.

Application

Anti-Phospho-PRAS40 (pThr246) antibody produced in rabbit has been used in western blotting.

Biochem/physiol Actions

Proline-rich Akt substrate of 40 kilodalton (PRAS40) acts as a linker between micro-environmental stress signaling and exosome-mediated secretion. The encoded protein might inhibit the activity of the mTOR complex 1 (mTORC1) kinase. Mutation in the gene is associated with the development of of cancers. Thus, this gene can be considered as a potential target for treatment of several cancers. PRAS40 plays a major role in the regulation of insulin sensitivity in human skeletal muscle cells (hSkMC).

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
