General description

Monoclonal Anti-ELMO1 (mouse IgG1 isotype) is derived from the hybridoma ELMO1-6 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide. Engulfment and cell motility 1 (ELMO1) protein is a member of the evolutionarily conserved family of ELMO proteins. The ELMO1 gene is mapped on the human chromosome at 7p14.2-p14.1.

Specificity

Monoclonal Anti-ELMO1 recognizes human and mouse ELMO1.

Immunogen

synthetic peptide corresponding to a sequence near the C-terminus of human ELMO1, conjugated to KLH. The corresponding sequence is identical in mouse, rat, bovine, monkey and dog and differs by 4 amino acids in ELMO2.

Application

Monoclonal Anti-ELMO1 antibody produced in mouse may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Engulfment and cell motility (ELMO) regulates the actin cytoskeleton reorganization during engulfment and cell migration. ELMO along with the dedicator of cytokinesis (Dock)-180 function as a guanine exchange factor (GEF) for the small guanosine triphosphatase (GTPase) Rac, leading to its activation and cytoskeleton rearrangement. ELMO1 interacts with brain-specific angiogenesis inhibitor 1 (BAI1), an engulfment receptor for apoptotic cells upstream of the ELMO1/Dock180/Rac module. High expression levels of ELMO1 and Dock180 are linked to the invasive phenotype of glioma cells.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage, freeze at -20oC in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
